Ins-1e Cells

INS-1E cells are an insulin-secreting rat insulinoma cell line used as a laboratory model of pancreatic beta-cell function, making them valuable for diabetes and metabolic research. In response to glucose, cellular metabolism increases the ATP-to-ADP ratio, which closes ATP-sensitive potassium channels, depolarizes the plasma membrane, and opens voltage-gated calcium channels; the resulting calcium influx triggers insulin-granule exocytosis. Researchers use INS-1E cells to study glucose-stimulated insulin secretion, beta-cell signaling, oxidative stress, and mechanisms of beta-cell dysfunction. They also support screening and evaluation of compounds intended to improve insulin secretion or protect beta cells.

Transcellular protein interactions are important determinants of pancreatic beta-cell function. Detailed here is a method—adapted from a coculture model of synaptogenesis—for investigating how specific transmembrane proteins influence insulin secretion. Transfected HEK293 cells express proteins of interest; beta cells do not need to be transfected or otherwise directly perturbed.

Darkfield Microscopy to Visualize Diffusional Dynamics of Nanorods on Cell Membrane

In this video, we describe the darkfield microscopy (DFM) technique combined with single-particle tracking (SPT) analysis to monitor the interaction of plasmonic gold nanorods with the cell membrane of a specific cell line of interest. Combined DFM and SPT analysis allows the visualization of the nanorod movement through the cell membrane with high spatiotemporal resolution.

Pancreatic cancer stem cells (CSCs) can be expanded in vitro using the anchorage-independent sphere culture technique, which represents a powerful tool to study CSC biology and can serve as the first step to develop novel CSC-targeting therapies. Here the methodology for expanding, analyzing and targeting of pancreatic CSCs is provided.
