Masson Trichrome

Masson trichrome is a histological staining technique that differentiates connective tissue from muscle and other cellular components, making tissue architecture and fibrosis easier to evaluate in medicine. The method applies sequential dyes, typically including iron hematoxylin, acid fuchsin, and aniline blue or light green, which produce contrasting colors in cell nuclei, cytoplasm, muscle, and collagen fibers. By highlighting collagen deposition, Masson trichrome supports the assessment of scarring, fibrosis, and structural changes in organs such as the liver, heart, and kidneys. It remains a valuable method for teaching histology, analyzing pathological specimens, and validating findings from modern imaging or molecular approaches.

Intrauterine adhesions (IUAs), the most common pathological consequence of endometrial injury, severely impair reproductive function. Trauma and infection are widely recognized as key risk factors for IUA development. We have successfully established a reproducible rat model of IUAs by combining two critical injury methods: mechanical curettage and infectious induction with lipopolysaccharide.

The post-mortem assessment of myocardial infarction (MI) in rodents is based on quantification of the infarct on stained heart sections. We describe an accurate method to quantify the infarct size using systematic sampling of harvested rat hearts from base to apex and image analyses of trichrome-stained histological sections.

A step-by-step video protocol of apical resection is demonstrated in this study. Apical resection is a recently highlighted surgical approach in mammalian heart regeneration research. This study may promote the application of apical resection as a standard methodology in research into the mechanism underlying heart regeneration.
