Single Cell Tracking

Single-cell tracking is a quantitative method for following individual cells over time, revealing behaviors that population-level measurements can conceal. Using time-lapse imaging, image analysis identifies cells in successive frames and links their positions to reconstruct trajectories, allowing researchers to measure migration, division, survival, and changes in cell state. In medicine, this approach helps characterize tumor-cell movement, immune-cell responses, infection dynamics, and responses to drugs while accounting for cellular heterogeneity. By connecting cell behavior with treatment conditions or disease progression, single-cell tracking supports more precise disease models, biomarker discovery, and evaluation of therapeutic strategies.

Source: Cabeen, M. T., et al. Single-cell Microfluidic Analysis of Bacillus subtilis. J. Vis. Exp. (2018)The video demonstrates the use of a microfluidic device to observe bacterial growth at the single-cell level. It showcases how non-motile bacterial cells, confined within micro-scale trenches, are continuously nourished and imaged under a fluorescence microscope. The setup enables real-time visualization of cell division and lineage tracking across multiple generations.
