Behavioral Stroke Indicators

Behavioral stroke indicators are sudden changes in cognition, communication, emotion, or goal-directed behavior that may signal disruption of brain function during a stroke. Reduced blood flow or bleeding damages neural circuits, producing signs such as confusion, difficulty speaking or understanding language, unusual behavior, impaired judgment, or spatial neglect, sometimes without obvious weakness. In neuroscience, recognizing these changes helps connect behavioral symptoms with affected brain regions and networks. Clinicians and caregivers can use them alongside physical warning signs to support rapid stroke recognition, neurological assessment, emergency treatment, and rehabilitation planning.

Here, we present a protocol to quantify nest building behavior in mice, which is known to be impaired in several neurological disorders and diseases. This protocol examines the utility of four materials and offers the opportunity to quantify the rater agreement in scoring, improving the validity and reliability of the assay.

Certain organic substances change color in dilute solution when the hydronium ion concentration reaches a particular value. For example, phenolphthalein is a colorless substance in any aqueous solution with a hydronium ion concentration greater than 5.0 × 10−9 M (pH < 8.3). In more basic solutions where the hydronium ion concentration is less than 5.0 × 10−9 M (pH > 8.3), it is red or pink. Substances such as phenolphthalein, which can be used to determine the pH of a solution, are called...

Electroencephalography Network Indices as Biomarkers of Upper Limb Impairment in Chronic Stroke

2023

The experimental protocol demonstrates the paradigm for acquiring and analyzing electroencephalography (EEG) signals during upper limb movement in individuals with stroke. The alteration of the functional network of low-beta EEG frequency bands was observed during the movement of the impaired upper limb and was associated with the degree of motor impairment.

A system for acquiring data from self-initiated individual behavior sessions within a social colony cage setting is presented. The efficacy of this system is demonstrated using an automated skilled reach assessment, enabling the characterization of post-stroke motor impairments, potential behavioral alterations related to motivation, circadian variations, and other innovative dependent variables.

Compensatory Limb Use and Behavioral Assessment of Motor Skill Learning Following Sensorimotor Cortex Injury in a Mouse Model of Ischemic Stroke

Mouse models have become increasingly popular in studies of behavioral neuroscience. As models advance, it is important to develop sensitive behavioral measures specific to the mouse. This protocol describes the Pasta Matrix Reaching Task, which is a skilled motor task for use in mouse models of stroke.
