Motor Neuron Disease Pathology

Motor neuron disease pathology is the study of the cellular, molecular, and anatomical changes that damage upper and lower motor neurons, the nerve cells that control voluntary movement. Degeneration in corticospinal pathways, spinal anterior horn cells, and brainstem motor nuclei disrupts signaling from the brain to skeletal muscle, producing progressive weakness, atrophy, and impaired movement. Abnormal protein accumulation, excitotoxic stress, mitochondrial dysfunction, and neuroinflammation can contribute to neuronal injury, although their relative roles vary across disease forms. Characterizing these mechanisms supports diagnosis, disease classification, biomarker development, and the search for treatments that preserve motor function.

Extracellularly Identifying Motor Neurons for a Muscle Motor Pool in Aplysia californica

In animals with large identified neurons (e.g. mollusks), analysis of motor pools is done using intracellular techniques1,2,3,4. Recently, we developed a technique to extracellularly stimulate and record individual neurons in Aplysia californica5. We now describe a protocol for using this technique to uniquely identify and characterize motor neurons within a motor pool.

We developed a new protocol to improve efficiency of in vitro differentiation of mouse embryonic stem cells into motor neurons. The differentiated ES cells acquired motor neurons features as evidenced by expression of neuronal and motor neuron markers using immunohistochemical techniques.
