Optical Window Installation

Optical window installation is a neurosurgical technique that places a transparent, sealed implant over exposed brain tissue, creating repeated visual access for in vivo study of neural structure and activity. Typically, a small section of skull is removed, the optical window is positioned over the target region, and the opening is stabilized and closed to maintain a clear imaging path while protecting the tissue. This preparation supports longitudinal microscopy, including fluorescence-based observation of neuronal dynamics, blood flow, and cellular responses. It enables researchers to relate neural activity to behavior and monitor changes across disease models or experimental interventions.

Installation of Optical Windows in the Brain of Transgenic Mice to Image Neural Activities

2025

This video demonstrates the brain's neural activity recording through an optical window in transgenic mice. The mice express a fluorescent protein that enables the detection of neural activities. A small portion of the skull is carefully removed to install an optical window, and the skull is then covered with black cement to block light before imaging.

Optical-resolution photoacoustic microscopy (OR-PAM) is an emerging technology capable of imaging optical absorption contrasts in vivo with cellular resolution and sensitivity. Here, we provide a visualized instruction on the experimental protocols of OR-PAM, including system configuration, system alignment, typical in vivo experimental procedures, and functional imaging schemes.
