Antibody Mediated Reactions

Antibody-mediated reactions are immune responses in which antibodies recognize and bind specific antigens, including drugs or drug-modified proteins, producing effects that can influence pharmacological safety and efficacy. Their mechanisms depend on the antibody class and target: IgE can cross-link on mast cells and trigger mediator release, while IgG or IgM may activate complement, promote phagocytosis, or form immune complexes that deposit in tissues. In pharmacology, these reactions help explain allergic and immune-related adverse drug reactions, guide evaluation of drug hypersensitivity, and support safer decisions about treatment selection, monitoring, and re-exposure.

Measuring the Activation of Fc-Mediated Effector Functions by HA Antibodies

2025

This video demonstrates an assay measuring the antibody-mediated activation of T cell effector functions. Adding an influenza virus hemagglutinin (HA)-specific monoclonal antibody to transfected mammalian cells expressing HA results in the formation of immune complexes with HA. Upon introducing engineered T cells expressing Fc receptors and a luciferase reporter, the T cells are activated by the antibody-HA complexes, and this activation is detected by adding a luciferase substrate and...

A Serum Bactericidal Assay for the Complement-Mediated Bactericidal Activity of Antibodies

2025

This video demonstrates a technique to evaluate the complement-mediated bactericidal activity of antibodies present in serum. This approach combines isolated serum with pathogenic bacteria and exogenous complement proteins. Upon incubation, the bacteria are grown on an agar plate, and the produced colonies are quantified to measure the serum bactericidal titer.

An Assay for Quantifying Antibody-Mediated Phagocytosis of Parasite-Infected Erythrocytes

This video demonstrates a flow cytometry-based phagocytosis assay involving labeled and opsonized Plasmodium falciparum-infected erythrocytes by monocytes. Flow cytometry identifies fluorescence in monocytes, confirming the successful identification of opsonized erythrocytes through phagocytosis.

A Method to Assess Fc-mediated Effector Functions Induced by Influenza Hemagglutinin Specific Antibodies

We describe a method to measure the activation of Fc-mediated effector functions by antibodies that target the influenza virus hemagglutinin. This assay can also be adapted to assess the ability of monoclonal antibodies or polyclonal sera targeting other viral surface glycoproteins to induce Fc-mediated immunity.

Overview Antibodies, also known as immunoglobulins (Ig), are essential players of the adaptive immune system. These antigen-binding proteins are produced by B cells and make up 20 percent of the total blood plasma by weight. In mammals, antibodies fall into five different classes, which each elicits a different biological response upon antigen binding. The Y-Shaped Structure of Antibodies Consists of Four Polypeptide Chains Antibodies consist of four polypeptide chains: two identical heavy...
