Nadph P450 Reductase

NADPH P450 reductase is a flavoprotein electron-transfer enzyme that supplies reducing equivalents to cytochrome P450 enzymes, making it central to oxidative metabolism and pharmacology. It binds NADPH and passes electrons sequentially through its FAD and FMN cofactors, enabling P450-dependent activation of molecular oxygen and oxidation of substrates. This partnership governs the biotransformation of many drugs, environmental chemicals, and endogenous compounds, influencing clearance, efficacy, toxicity, and drug interactions. Studying NADPH P450 reductase helps researchers interpret metabolic pathways and assess how changes in reductase activity can alter pharmacological responses.

NADPH oxidase is the major source of reactive oxygen species (ROS) in phagocytes. Because of the ephemeral nature of ROS, it is difficult to measure and monitor ROS levels in living animals. A minimally invasive method for serial quantification of ROS in living mice is described.
