Un protocole détaillé est fourni pour l’utilisation de la technologie CRISPR/Cas9 afin d’obtenir un knock-in ciblé très efficace de grandes constructions multicistroniques dans les cellules T humaines primaires via la voie de réparation de l’ADN HMEJ (Homology-Mediated End Joindre). Les lymphocytes T conçus avec ce protocole adaptable à la GMPc maintiennent une excellente expansion cellulaire, une cytotoxicité et une production de cytokines.
