JoVE Neuroscience is a multidisciplinary section devoted to investigations of the structure, function, physiology, and pathophysiology of the brain and nervous system. JoVE Medicine serves as a connection between basic research and clinical applications. Articles may present medical procedures, case studies or clinical trial methodologies. JoVE Engineering encompasses a broad range of experimental and instrumental techniques utilized in physics research. Investigations in this area strive to address and answer a broad range of scientific questions, such as device mechanisms and efficiencies, using physical tools. This approach often requires a combination of specialties, and research in this area tends to be interdisciplinary with contributions from mechanical, electrical, and chemical engineers.

Ubiquitination is a critical regulator of intracellular protein functions. Here, we describe two cell lysis methods for detecting substrate protein ubiquitination levels in mammalian cells and compare their effectiveness in analyzing ubiquitination levels. Protein overexpression in HEK293T cells was induced by transient transfection, and ubiquitinated proteins were detected by western blot analysis.

This protocol describes how to construct and operate a small, low-cost growth chamber capable of maintaining extreme carbon dioxide (exCO2) levels for plant experiments.
