A distinct subclass of glycoproteins called proteoglycans contains repeating units of disaccharides that form long-chain sugars called glycosaminoglycans or GAGs.
GAGs are negatively charged and remain surrounded by water molecules, creating swelling pressure within tissues. This allows the tissues to withstand compressional forces.
Proteoglycans are synthesized in the Golgi when galactosamine transferase transfers N-acetylgalactosamine from UDP-N- galactosamine to a serine or threonine on the polypeptide in O-type glycosylation.
As the protein moves within the Golgi lumen, glycosyltransferase adds other functional units such as galactose, sialic acid, N-acetylglucosamine, or fucose. Finally, the fully formed proteoglycan is released in secretory vesicles to the cytosol.
