Question 1

What is the basal lamina and how does it differ from the general extracellular matrix?

Accepted Answer

The basal lamina is a specialized form of extracellular matrix, a thin layer lying beneath cells that separates them from other tissues. Unlike the general ECM, the basal lamina has a defined three-layer structure: lamina lucida, lamina densa, and lamina reticularis. It serves specific functions depending on location, such as filtering in the kidney or providing structural support at the neuromuscular junction.

Question 2

What are the main protein components that make up the basal lamina?

Accepted Answer

The basal lamina is primarily composed of type IV collagen and glycoproteins including laminins, entactin, and perlecan. These proteins work together to form a structural framework that provides attachment sites for epithelial cells. The laminins are the adhesive proteins of basal lamina that facilitate cell-matrix interactions and bridge connections between cells and the surrounding connective tissue.

Question 3

How does the basal lamina function in the kidney glomerulus?

Accepted Answer

In the kidney glomerulus, the basal lamina is positioned between sheets of endothelial and epithelial cells, where it acts as a molecular filter. This specialized filtration barrier prevents macromolecules from the blood from passing into the urine while allowing smaller molecules and water to cross, maintaining proper kidney function and preventing protein loss.

Question 4

What role does the basal lamina play at the neuromuscular junction?

Accepted Answer

At the neuromuscular junction, the basal lamina surrounds muscle cells and separates them from nerve cells at the synapse. Beyond structural separation, it aids in localizing acetylcholine receptors on the muscle membrane and helps regenerate the synapse after injury, making it essential for proper neuromuscular transmission and recovery.

Question 5

How do cells attach to the basal lamina?

Accepted Answer

Cells attach to the basal lamina through cell membrane proteins like integrins and dystroglycan, which bind to basal lamina proteins such as laminins and fibronectin. This interaction creates a bridge that associates cells with the extracellular matrix of the connective tissue, providing structural stability and facilitating cell-matrix communication.

Question 6

How does the basal lamina separate different tissue types?

Accepted Answer

The basal lamina functions as a selective barrier between different tissue types. In epithelial tissues, it lies below the epithelial cells, separating them from underlying connective tissue. Its organization and specific protein composition depend on the cell type it supports, allowing it to maintain tissue integrity while controlling molecular exchange between adjacent tissue compartments.

Question 7

What makes type IV collagen important to basal lamina structure?

Accepted Answer

Type IV collagen is the major structural component of the basal lamina, providing the primary framework for the three-layer structure. Unlike fibrillar collagens found elsewhere in the ECM, type IV collagen forms a mesh-like network that, combined with glycoproteins, creates the stable, selective barrier essential for basal lamina function across different tissues.
