Apoptotic Platelets

Apoptotic platelets are anucleate blood cells that undergo an apoptosis-like form of programmed cell death, a process important for regulating platelet lifespan and hemostasis. Intrinsic cellular stress can disrupt mitochondrial membrane potential, activate caspases, and promote phosphatidylserine exposure on the platelet surface, marking these cells for recognition and clearance. In biochemistry, studying apoptotic platelets helps clarify how mitochondrial signaling, proteolysis, membrane remodeling, and phagocytic removal control platelet turnover. These mechanisms are relevant to thrombocytopenia, inflammation, cardiovascular disease, and the development of platelet-based diagnostic assays or therapies.

Turbidimetry on Human Washed Platelets: The Effect of the Pannexin1-inhibitor Brilliant Blue FCF on Collagen-induced Aggregation

We describe a straightforward method for the isolation of washed platelets from human blood followed by agonist-induced platelet aggregation measurements by turbidimetry. As an example we apply this method for studying the aggregation response of human platelets to collagen after a pre-incubation with the Pannexin1 channel inhibitor Brilliant Blue FCF.

In this video, we describe a protocol for purifying platelets from murine whole blood and their subsequent activation with the platelet agonist thrombin. When analyzed with flow cytometry, the purified resting platelets are characterized based on the expression of their cell-surface marker CD41, whereas purified activated platelets co-express both the surface markers CD41 and CD62P.
