Biotin-lambda-dna Immobilization

Biotin-lambda-DNA immobilization is a biochemical technique that anchors bacteriophage lambda DNA to a functionalized surface, enabling controlled observation and manipulation of individual DNA molecules. In this approach, biotin attached to a DNA terminus binds with high affinity to streptavidin or avidin on the surface, while solution conditions and surface chemistry help preserve the DNA’s accessibility and stability. The resulting tether supports fluorescence imaging, force measurements, and assays of DNA-binding proteins, enzymes, and molecular motors. By converting freely diffusing lambda DNA into a surface-associated substrate, the method facilitates single-molecule studies of DNA structure, dynamics, and protein-mediated processes.

The reversible addition of palmitate to proteins is an important regulator of intracellular protein trafficking. This is of particular interest in neurons where many synaptic proteins are palmitoylated. We utilize a simple biochemical method to detect palmitoylated proteins in cultured neurons, which can be adapted for multiple cell types and tissues.
