High-throughput Purification

High-throughput purification is a biochemistry strategy for isolating many proteins or other biomolecules in parallel, reducing the time and labor required for sample preparation. It combines automated liquid handling with miniaturized, parallel workflows in which binding, washing, and elution occur across multiple samples, often using affinity, ion-exchange, or size-based chromatography. Researchers can rapidly compare purification conditions, process protein variants, and generate consistent material for biochemical assays, structural studies, and drug discovery. By increasing experimental capacity while conserving samples and reagents, high-throughput purification supports systematic screening and accelerates the development of reproducible purification protocols.

Plasmid purification is a technique used to isolate and purify plasmid DNA from genomic DNA, proteins, ribosomes, and the bacterial cell wall. A plasmid is a small, circular, double-stranded DNA that is used as a carrier of specific DNA molecules. When introduced into a host organism via transformation, a plasmid will be replicated, creating numerous copies of the DNA fragment under study. In this video, a step-by-step generalized procedure is described for how to perform plasmid purification.
