Ni-nta Resin Purification

Ni-NTA resin purification is a form of immobilized metal affinity chromatography used to isolate recombinant proteins, particularly those carrying a polyhistidine tag. Nickel ions coordinated by nitrilotriacetic acid on the resin bind histidine residues, allowing tagged proteins to remain retained while unbound cellular components are removed during washing. The target protein is typically eluted by increasing imidazole concentration, which competes with histidine for nickel binding, or by changing pH under suitable conditions. This method provides a selective and efficient step for protein production, biochemical characterization, structural studies, and downstream assays.

Flow Cytometry-based Purification of S. cerevisiae Zygotes

To purify zygotes of S. cerevisiae, haploid cells of opposite mating type were engineered to express red or green fluorescent proteins, co-incubated to allow zygote formation, and fractionated using a flow cytometry-based protocol. The highly-enriched fraction enables subsequent "-omic" studies, recovery of initial progeny, and systematic investigation of zygote morphogenesis.

A method is described for efficient purification of twin-Strep-tagged fusion proteins and their specific complexes on modified streptavidin (Strep-Tactin) resin covalently cross-linked with Bis(sulfosuccinimidyl) suberate (BS3). The method has the advantages of fast speed, good target protein recovery and high purity, and is compatible with subsequent analysis by mass spectrometry.

The synthesis of asymmetric species of ferrocene is challenging using solution techniques. This report focuses on the methods carried out to produce a ferrocene-biotin bioconjugate using facile and clean reactions accomplished via solid-phase synthesis.
