Uhplc Msms Analysis

UHPLC-MS/MS analysis is an analytical method that combines ultra-high-performance liquid chromatography with tandem mass spectrometry to separate, identify, and quantify chemical compounds in complex samples. UHPLC first resolves molecules by differences in interactions with a chromatographic column, while mass spectrometry ionizes them and measures mass-to-charge ratios; selected precursor ions are then fragmented, and their product ions provide compound-specific signals. In biochemistry, this approach supports sensitive analysis of metabolites, lipids, peptides, proteins, and drug-related compounds in biological samples. Its high selectivity and quantitative capability make it valuable for pathway studies, biomarker research, and investigating molecular changes in health and disease.

The present method serves to identify isoform-specific inhibitors of histone deacetylases (HDAC) in HeLa cells by the UHPLC-MS analysis of multiple substrates. This is an antibody-free method developed to reflect HDAC1 and HDAC6 activity in the living cell environment, in contrast to single-isoform cell-free assays.

A specific and sensitive method to gain insight into the expression profile of glycosphingolipid antigens in immune organs and cells is described. The method takes advantage of the ion trap mass spectrometry allowing step-wise fragmentation of glycosphingolipid molecules for structural analysis in comparison to chemically synthesized standards.

A Neuronal and Astrocyte Co-Culture Assay for High Content Analysis of Neurotoxicity

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Results demonstrate that high content analysis represents an exciting novel technology for neurotoxicity assessment.
