Vesicle Characterization

Vesicle characterization is the systematic analysis of membrane-bound vesicles to determine their physical, chemical, and biological properties, supporting reliable interpretation of their roles in biochemistry. Researchers combine methods such as dynamic light scattering or nanoparticle tracking analysis to measure size and concentration, microscopy to assess morphology, and biochemical assays to examine lipid composition, proteins, and molecular cargo. Surface-marker analysis can further distinguish vesicle populations and evaluate sample purity. These measurements are essential for studying vesicle formation, transport, and signaling, while improving reproducibility in research involving extracellular vesicles, drug delivery systems, and other nanoscale membrane structures.

This protocol details a method to isolate extracellular vesicles (EVs), small membranous particles released from cells, from as little as 10 μl serum samples. This approach circumvents the need for ultracentrifugation, requires only a few minutes of assay time, and enables the isolation of EVs from samples of limited volumes.
