Immunocompetent Rats

Immunocompetent rats are laboratory rats with intact, functioning immune systems, making them valuable models for studying how engineered materials, devices, and tissues interact with a complete host response. Their immune cells can recognize implanted biomaterials and trigger inflammation, foreign-body reactions, vascularization, tissue repair, or rejection, depending on the material and experimental context. In bioengineering, these models support evaluation of biocompatibility, drug delivery systems, tissue-engineered constructs, implants, and regenerative therapies under physiologically relevant conditions. Because they preserve immune interactions absent from immunodeficient models, immunocompetent rats can help reveal both therapeutic effects and safety limitations before further translational development.

GL261 glioma cells provide a useful immunocompetent animal model of glioblastoma. The goals of this protocol are to demonstrate proper techniques for monitoring intracranial tumor growth using in vivo bioluminescence imaging, and to verify the utility of luciferase-modified GL261 cells for studying tumor immunology and immunotherapeutic approaches for treating glioblastoma.

A method for establishing lung infections in immunocompetent rodents is described. With proficiency, this method can be performed quickly and easily to induce stable infection with many isolates of S. pneumoniae, H. influenzae, P. aeruginosa, K. pneumoniae and A. baumannii.
