Sialic Acid Modification

Sialic acid modification is the enzymatic or chemical alteration of sialic acid residues, sugars commonly found at the ends of cell-surface glycans and glycoproteins. In bioengineering, these changes can adjust glycan composition through sialyltransferase-mediated addition, sialidase-mediated removal, or metabolic and chemical engineering of sialic acid pathways. Because terminal sialylation influences molecular recognition, stability, charge, and interactions with cells, researchers use this approach to design glycoproteins, biomaterials, and engineered cell systems. Controlled sialic acid modification supports the development of biopharmaceuticals, targeted delivery platforms, and experimental models for investigating cell signaling and host-pathogen interactions.

Sialic acid is a typical monosaccharide-unit found in glycoconjugates. It is involved in a plethora of molecular and cellular interactions. Here we present a method to modify cell surface sialic acid expression using metabolic glycoengineering with N-acetylmannosamine derivatives.
