Syringe Pump Perfusion

Syringe pump perfusion is a controlled fluid-delivery technique that uses a syringe pump to move culture medium or other solutions through a biological system at a defined flow rate. A motor-driven mechanism advances the syringe plunger, generating steady, programmable flow through tubing, channels, scaffolds, or tissue-engineered constructs. In bioengineering, this approach supports continuous nutrient delivery, waste removal, and controlled exposure to biochemical or mechanical cues in cell culture, bioreactors, and microfluidic platforms. By improving control over the cellular environment, syringe pump perfusion helps researchers study tissue development, evaluate biomaterials, and design more physiologically relevant in vitro models.

Here we present a protocol to construct a pressure-controlled syringe pump to be used in microfluidic applications. This syringe pump is made from an additively manufactured body, off-the-shelf hardware, and open-source electronics. The resulting system is low-cost, straightforward to build, and delivers well-regulated fluid flow to enable rapid microfluidic research.

In this video, we perform Langendorff retrograde perfusion of an isolated mouse heart. During the procedure, the cardiac activity of the heart is maintained by perfusing it retrogradely with a perfusion solution via the coronary arteries using a cannula inserted into the ascending aorta. The model is used to study the cardiac pathophysiology of the heart.
