Thermogenic Gene Program

The thermogenic gene program is a coordinated set of gene-expression changes that enables cells, especially brown and beige adipocytes, to produce heat rather than store energy. Its core mechanism involves transcriptional activation of thermogenic factors such as uncoupling protein 1 (UCP1), which dissipates the mitochondrial proton gradient and releases energy as heat instead of converting it into ATP. In bioengineering, researchers use this program to design metabolically active adipose tissues, develop cell-based models of energy metabolism, and investigate strategies for regulating body weight and metabolic disease. Understanding its regulatory network also supports efforts to engineer tissues with improved physiological function.

Programmed electrical stimulation provides the ability to determine conduction properties of the heart, and the possibility to induce and terminate cardiac arrhythmias using various pacing protocols. Using a transvenous catheter, intracardiac electrogram recordings can be obtained in mice following programmed electrical stimulation protocols to identify arrhythmogenic substrates.

Programming Stem Cells for Therapeutic Angiogenesis Using Biodegradable Polymeric Nanoparticles

We describe the method of programming stem cells to overexpress therapeutic factors for angiogenesis using biodegradable polymeric nanoparticles. Processes described include polymer synthesis, transfecting adipose-derived stem cells in vitro, and validating the efficacy of programmed stem cells to promote angiogenesis in a murine hindlimb ischemia model.
