Human Ige Fc-receptors

Human IgE Fc-receptors are cell-surface proteins that bind the constant Fc region of immunoglobulin E and regulate allergic and immune responses. The high-affinity receptor FcεRI is expressed mainly on mast cells and basophils, where allergen-mediated cross-linking of receptor-bound IgE activates tyrosine kinases, receptor phosphorylation, and release of inflammatory mediators; the low-affinity receptor FcεRII, or CD23, influences IgE transport, uptake, and immune regulation. Studying these receptors with flow cytometry, binding assays, and antibody-based methods helps characterize receptor expression, signaling, and ligand interactions. These techniques support research on allergy, asthma, host defense, therapeutic antibodies, and strategies that target IgE-dependent inflammation.

In vitro analysis of class switch recombination in mice is challenging due to cytophilic IgE molecules bound to Fc receptors on the surface of B cells. We describe a method for IgE detection using trypsin-mediated cleavage of surface-bound IgE prior to fixation and permeabilization for cytoplasmic fluorescence staining.

Basophil activation test is a potent tool for the detection of IgE-dependent allergies in vitro. Here, an optimized protocol for basophil activation test is used to investigate drug hypersensitivity. A method for the efficient production of covalent drug-protein conjugates and their physicochemical characterization is described.

Compared with traditional affinity chromatography using protein A agarose bead-packed columns, protein A membrane adsorbers can significantly speed laboratory-scale isolation of antibodies and other Fc fragment-expressing proteins. Appropriate analysis and quantification methods can further accelerate protein processing, allowing isolation/characterization to be completed in one workday, instead of 20+ work hours.
