Mediator Release Assay

A mediator release assay is a biological technique that measures signaling molecules secreted by activated cells, providing a functional readout of cellular responses to a stimulus. In a typical assay, cells are exposed to a test substance or control, and activation triggers the release of mediators into the surrounding fluid; researchers then quantify these molecules using biochemical or immunological detection methods. The approach is used to assess immune-cell activation, characterize inflammatory responses, evaluate allergic sensitization, and investigate drug effects or toxicity. By linking a controlled stimulus to measurable mediator production, the assay supports mechanistic studies and comparative analysis of cellular function.

Here we present the mediator release assay, using a rat basophilic leukemia cell line transfected with the human IgE receptor, to simulate the degranulation of effector cells typically observed in type 1 allergic reactions. This method investigates the biological activity of allergens in a highly sensitive, reproducible, and tailorable manner.

The antimicrobial protein degrades the bacteria's dye-labeled cell wall and releases the dye molecules turning the solution blue. The absorbance of the solution correlates with the enzymatic activity of the antimicrobial protein.
