Nadh Absorbance

NADH absorbance is the measurement of light absorbed by reduced nicotinamide adenine dinucleotide, a coenzyme central to cellular electron transfer and a useful indicator of biochemical reactions. NADH strongly absorbs ultraviolet light at approximately 340 nm, whereas its oxidized form, NAD+, absorbs much less at this wavelength, allowing changes in absorbance to track NADH production or consumption. In biological techniques, researchers use spectrophotometers to monitor enzyme activity, reaction kinetics, and redox transformations in real time. This approach supports studies of metabolism, dehydrogenase function, and energy-related pathways, while providing a quantitative, noninvasive readout of molecular change.

The objective is to monitor the mitochondrial redox state of isolated hearts within the context of physiologic preload and afterload pressures. A biventricular working rabbit heart model is presented. High spatiotemporal resolution fluorescence imaging of NADH is used to monitor the mitochondrial redox state of epicardial tissue.

Here we describe a method to directly visualize microregional tissue hypoxia in the mouse cortex in vivo. It is based on concurrent two-photon imaging of nicotinamide adenine dinucleotide (NADH) and the cortical microcirculation. This method is useful for high resolution analysis of tissue oxygen supply.
