Attenuation Mechanism

The attenuation mechanism is a form of gene regulation in which bacteria adjust transcription in response to cellular conditions, particularly the availability of specific amino acids. In many operons, a ribosome translates a leader peptide while RNA polymerase transcribes the adjacent leader sequence; changes in translation cause the RNA to fold into alternative secondary structures that either form a transcription terminator or permit transcription to continue. This mechanism links translation with transcription and enables rapid control of biosynthetic genes, as seen in the tryptophan operon. Studying attenuation clarifies how bacteria conserve resources and adapt gene expression to nutrient conditions.

Transcription Attenuation in Prokaryotes

2020

Transcriptional attenuation occurs when RNA transcription is prematurely terminated due to the formation of a terminator mRNA hairpin structure. Bacteria use these hairpins to regulate the transcription process and control the synthesis of several amino acids including histidine, lysine, threonine, and phenylalanine. Transcription attenuation takes place in the non-coding regions of mRNA. There are several different mechanisms used to attenuate transcription. In ribosome mediated...

2015

The goal of the protocol described in this paper is to induce compulsive-like behavior in rats for the study of obsessive-compulsive disorder (OCD). This behavior is precipitated by attenuating a signal indicating that a lever-press response was effective in producing food.

Intradermal Immunization with Live Attenuated Sporozoites in a Mouse Model

This video demonstrates an assay for immunizing a mouse model with live attenuated sporozoites. The radiation-attenuated Plasmodium sporozoites are injected into the dorsal side of the mouse's ear. After injections at multiple locations on both ears, the mouse is allowed to recover.

Generating Recombinant Live-Attenuated Influenza Viruses for Vaccines
