Cellular Immune Assays

Cellular immune assays are laboratory methods that measure how immune cells respond, communicate, and perform effector functions, making them essential for studying host defense and immune regulation. These assays typically expose isolated cells or mixed cell populations to antigens, mitogens, or target cells, then quantify responses such as proliferation, cytokine secretion, activation-marker expression, or cytotoxicity using methods including flow cytometry and ELISpot. In biology, cellular immune assays help characterize immune-cell function, compare responses in health and disease, evaluate vaccines and immunotherapies, and assess drug effects, providing functional evidence that complements molecular and serological measurements.

This video describes a colorimetric assay for quantitatively measuring pyruvate in the cellular fractions of Caenorhabditis elegans. Utilizing the hydrogen peroxide produced from the enzymatic reaction of pyruvate oxidase with cellular pyruvate, a chromogenic dye is oxidized to produce a colored product, measured using colorimetry to determine the pyruvate concentration.
