Crispr-cas9 Genome Editing

CRISPR-Cas9 genome editing is a molecular method for making targeted changes to DNA, enabling researchers to investigate gene function and potentially correct disease-associated variants. A guide RNA matches a chosen DNA sequence and directs the Cas9 nuclease to cut both DNA strands near an adjacent PAM sequence; the cell then repairs the break through error-prone non-homologous end joining or, when a repair template is supplied, homology-directed repair. In biology, this technology supports gene knockout, precise sequence modification, functional genomics, disease modeling, and studies of development. Its efficiency and programmability have also advanced research in agriculture, biotechnology, and therapeutic development.

2025

The CRISPR-Cas system serves as a bacterial defense mechanism against invading genetic elements such as viruses and plasmids, forming the foundation for its adaptation as a powerful genome-editing tool. Originally discovered in prokaryotes, this system has been repurposed to revolutionize genetic engineering across a wide range of organisms, including plants, animals, and humans. The core component, Cas9, is an endonuclease derived from Streptococcus pyogenes, capable of introducing...

A well-established technique for modifying specific sequences in the genome is gene targeting by homologous recombination, but this method can be laborious and only works in certain organisms. Recent advances have led to the development of “genome editing”, which works by inducing double-strand breaks in DNA using engineered nuclease enzymes guided to target genomic sites by either proteins or RNAs that recognize specific sequences. When a cell attempts to repair this damage, mutations can be...

In this video, we demonstrate all-in-one CRISPR-Cas9 based genome editing in cultured cells where Cas9 and sgRNA are provided as a single plasmid construct to the cells. The CRISPR-Cas9 system and desired gene to be inserted was introduced in cells through electroporation technique to facilitate successful gene editing.

A Technique for Gene Editing in Natural Killer Cells Using CRISPR Cas9

2025

This video demonstrates a technique for Cas9 ribonucleoprotein-mediated genetic modification of primary natural killer (NK) cells. A Cas9 ribonucleoprotein, consisting of a Cas9 endonuclease bound to a guide RNA (gRNA) formed by base pairing a CRISPR RNA (crRNA) and a trans-activating crRNA (tracrRNA), is introduced into primary natural killer cells via electroporation. The ribonucleoprotein targets and cleaves the host DNA at the target site, leading to gene knockout via modification of the...
