Parietal Pleura

The parietal pleura is the outer layer of the pleural membrane, a thin serous lining that surrounds each lung and lines the inner chest wall, diaphragm, and mediastinum. It secretes and contacts a small amount of pleural fluid, which reduces friction between the parietal and visceral pleura as breathing movements change the volume of the thoracic cavity; sensory nerves in this layer also contribute to pain perception. Understanding the parietal pleura is essential in biology and medicine because inflammation, air, or excess fluid in the pleural space can impair lung expansion and produce characteristic clinical signs.

The endothelial glycocalyx/endothelial surface layer is ideally studied using intravital microscopy. Intravital microscopy is technically challenging in a moving organ such as the lung. We demonstrate how simultaneous brightfield and fluorescent microscopy may be used to estimate endothelial surface layer thickness in a freely-moving in vivo mouse lung.
