Single Cycle Virus

A single-cycle virus is a viral system engineered or selected to complete only one round of infection, making it useful for studying viral biology while limiting further spread. Its genome retains the functions needed for cell entry, gene expression, and genome replication, but lacks an essential component for producing infectious progeny; this defect can be supplied during virus production in specialized complementing cells. In biology research, single-cycle viruses help separate viral entry from replication and transmission, evaluate antiviral responses, and develop safer vaccine or gene-delivery platforms. Their restricted life cycle also supports controlled analysis of host-virus interactions under laboratory conditions.

Single Virus Genomics (SVG) is a method to isolate and amplify the genomes of single virons. Viral suspensions of a mixed assemblage are sorted using flow cytometry onto a microscope slide with discrete wells containing agarose, thereby capturing the virion and reducing genome shearing during downstream processing. Whole genome amplification is achieved using multiple displacement amplification (MDA) resulting in genomic material that is suitable for sequencing.

What are Biogeochemical Cycles?

2019

The most common elements in organic molecules, carbon, hydrogen, oxygen, nitrogen, sulfur, and phosphorus, are only available in the ecosystem in limited amounts. Therefore, these nutrients must be recycled through both biotic and abiotic components of the ecosystem, in processes generally called biogeochemical cycles. Biogeochemical Cycles and Decomposition The matter that makes up living organisms, like water, carbon, nitrogen, sulfur and phosphorous, exist in limited quantities within the...

Cell Cycle Analysis

Cell cycle refers to the set of events through which a cell grows, replicates its genome, and ultimately divides into two daughter cells through the process of mitosis. Because the amount of DNA in a cell shows characteristic changes throughout the cycle, techniques known as cell cycle analysis can be used to separate a population of cells according to the different phases of cell cycle they are in, based on their varying DNA content. This video will cover the principles behind cell cycle...
