Viral Hepatitis

Viral hepatitis is inflammation of the liver caused by infection with hepatitis viruses, making it an important subject in biology, infectious disease, and public health. After entering the body, these viruses target hepatocytes and use host-cell machinery to replicate; the resulting immune response can damage liver tissue, while persistent infection may lead to fibrosis, cirrhosis, or liver cancer. Studying viral hepatitis clarifies host-pathogen interactions, transmission routes, and mechanisms of acute versus chronic disease. This knowledge supports diagnostic testing, vaccination, antiviral treatment, and prevention strategies that reduce transmission and long-term liver-related complications.

Suppressing Hepatitis B Virus Replication Using Viral Antigen-Specific T Cells

The video demonstrates an assay using human induced pluripotent stem cell (iPSC)-derived CD8+ T cells to inhibit hepatitis B virus (HBV) replication in mice. These T cells, engineered with HBV-specific receptors, are transferred into transgenic mice. Replication-competent HBV genome plasmids are then injected directly into the liver, enabling viral DNA uptake. Once viral antigens are expressed and presented by hybrid MHC-I molecules, the engineered T cells recognize and bind to these complexes.

We describe here the in vitro generation of HBV DNA via a Hepatitis B virus infection system and the highly sensitive detection of its (1–2 copies) integration using inverse nested PCR.
