Cryptococcal Antigen Testing

Cryptococcal antigen testing is a laboratory method for detecting capsular polysaccharide produced by Cryptococcus species, helping identify cryptococcosis before or alongside culture-based diagnosis. In commonly used lateral flow or agglutination assays, patient serum or cerebrospinal fluid is exposed to specific antibodies that bind the cryptococcal antigen, generating a visible test signal; antigen concentration can also be reported as a titer. The test supports rapid evaluation for cryptococcal meningitis and disseminated infection, particularly in people with advanced immunosuppression, and can guide confirmatory testing and clinical management. In biology, it illustrates how pathogen-specific molecular markers translate into practical diagnostic information.

2026

Cryptococcal meningitis is a life-threatening opportunistic infection predominantly associated with HIV/AIDS, accounting for over 100,000 deaths annually worldwide. However, it also affects individuals with other forms of immunosuppression, including those undergoing immunosuppressive therapy, organ transplant recipients, patients with innate immunodeficiencies, and individuals with hematological disorders. The infection is caused mainly by Cryptococcus neoformans and Cryptococcus gattii,...

In this antigen-driven colitis model, OT-II CD4+ T cells expressing a red fluorescent protein were adoptively transferred into RAG-/- mice that express a green fluorescent protein in mononuclear phagocytes (MPs). The hosts were challenged with Escherichia coli (E.coli) expressing the ovalbumin protein (OVA) fused to a cyan fluorescent protein (CFP).

This technique describes an automated batch image processor designed to measure polysaccharide capsule and body radii. While initially designed for Cryptococcus neoformans capsule measurements the automated image processor can also be applied to other contrast based detection of circular objects.

Extraction of Tissue Antigens for Functional Assays
