Radio Tlc Analysis

Radio TLC analysis, or radio thin-layer chromatography, is a technique for separating and measuring radiolabeled compounds on a thin adsorbent layer, making it useful for assessing chemical composition and radiochemical purity. A sample is applied to a silica or other stationary phase, developed with a suitable solvent, and separated according to differences in adsorption and solubility; the radioactive components are then located and quantified using autoradiography, phosphor imaging, or a radioactivity scanner. In chemistry, this method helps monitor radiochemical reactions, identify labeled products, detect impurities, and evaluate compound stability, supporting research in synthesis, tracer studies, and pharmaceutical development.

Thin-Layer Chromatography (TLC): Overview

2025

Thin-layer chromatography (TLC) is a chromatography technique that separates compounds based on their polarity. TLC typically uses polar silica gel, a form of silicon dioxide, as the stationary phase. The silica gel contains hydroxyl (OH) groups on its surface, which form hydrogen bonds with polar compounds, influencing their adhesion to the stationary phase. To begin the analysis, a mixture of compounds is spotted on the starting line on the TLC plate using a thin capillary. The bottom of the...

Composition of polar lipid extracts and the fatty acid composition of individual glycerolipids are determined in a simple and robust lipid profiling experiment. For this purpose, glycerolipids are isolated by thin layer chromatography and subjected to transmethylation of their acyl groups. Fatty acyl methylesters are quantified by gas-liquid chromatography.

Guanosine triphosphate (GTP) binding is one of the earliest events in G-Protein-Coupled Receptor (GPCR) activation. This protocol describes how to pharmacologically characterize specific GPCR-ligand interactions by monitoring the binding of the radio-labeled GTP analog, [35S]guanosine-5'-O-(3-thio)triphosphate ([35S]GTPγS), in response to a ligand of interest.
