Preclinical Therapeutic Testing

Preclinical therapeutic testing is the evaluation of a potential treatment’s safety, biological activity, and therapeutic effects before it is studied in humans. Researchers test compounds or biological interventions in cell cultures, organoids, and animal models, measuring dose responses, molecular targets, tissue effects, toxicity, and developmental outcomes under controlled conditions. In developmental biology, these studies can reveal how therapies influence cell differentiation, tissue formation, disease mechanisms, and recovery from developmental defects. The resulting evidence helps prioritize promising candidates, refine dosing and delivery strategies, identify risks, and determine whether a treatment is sufficiently supported for clinical research.

This protocol provides methods for generating 3D engineered cardiac and skeletal muscle tissues and describes their use in preclinical drug screening modalities. The described methods utilize a magnetic sensing system to facilitate the simultaneous assessment of 24 tissues in parallel.

A method for reliably grafting luciferase-tagged human malignant peripheral nerve sheath tumor cells into the sciatic nerve of immunodeficient mice is described. The use of bioluminescence imaging to demonstrate proper establishment of tumor grafts and criteria for random segregation of animals into study groups are also discussed.

Established cancer cell lines and xenografts have been the mainstay of cancer research for the past several decades. However, recent evidence suggests that therapeutic response is greatly influenced by the tumor cell microenvironment. Therefore, we have developed an ex vivo analysis of primary tumor specimens for drug development purposes.
