Selective Medium Culture

Selective medium culture is a laboratory method that uses a specially formulated growth medium to favor particular cells or microorganisms while restricting others, making it useful for isolating defined populations. Its action depends on adjusting nutrients, signaling factors, or selective agents so that target cells meet their growth requirements, whereas unwanted cells are inhibited or fail to survive. In developmental biology, this approach helps enrich progenitor or differentiated cell populations and maintain them under controlled conditions for studying cell fate, lineage progression, and tissue formation. Selective cultures can improve experimental consistency and support the development of models for investigating developmental mechanisms.

Blaize1, Elizabeth Suter1 1 Department of Biological Sciences, Wagner College, 1 Campus Road, Staten Island NY, 10301 Prokaryotic cells are able to inhabit nearly every environment on this planet. As a kingdom, they possess a great metabolic diversity, allowing them to use a wide variety of molecules for energy generation (1). Therefore, when cultivating these organisms in the lab, all necessary and specific molecules required to make energy must be...

Described here is a protocol for the determination of four different tryptophan metabolites generated in the kynurenine pathway (kynurenine, 3-hydroxykynurenine, xanthurenic acid, 3-hydroxyanthranilic acid) in the medium collected from cancer cell cultures analyzed by liquid chromatography coupled with a single quadrupole mass spectrometry.

Selectively Removing Microglia from a Rat Cerebellar Granule Neuron Culture

This video demonstrates a technique for the selective removal of microglia from a primary rat neuronal culture. Cerebellar granule neurons are treated with an antimetabolite to eliminate proliferating glial cells. Additionally, they are treated with lysosome-targeting agents to deplete microglia from the culture.
