Transgenic Fish Embryos

Transgenic fish embryos are early-stage fish embryos whose genomes contain an introduced DNA sequence, making them useful for studying how genes regulate development in a living organism. Researchers commonly inject DNA constructs into fertilized eggs, where the sequence may integrate into the genome and drive expression through a selected promoter; fluorescent reporter genes can then reveal when and where the transgene is active. In developmental biology, these embryos support analysis of cell fate, tissue formation, gene function, and developmental signaling through direct observation over time. They also provide models for testing genetic hypotheses and linking molecular changes to visible developmental outcomes.

In this video, we will demonstrate how to record electrical activity from identified single neurons in a whole brain preparation, which preserves complex neural circuits. We use transgenic fish in which gonadotropin-releasing hormone (GnRH) neurons are genetically tagged with a fluorescent protein for identification in the intact brain preparation.

This video demonstrates the ex vivo culture and imaging of oculomotor slices from transgenic mouse embryos. The embryos are isolated and embedded in agarose. Using a vibratome, slices containing the oculomotor nuclei and eyes are obtained. The slices are maintained in culture and imaged using a fluorescence microscope.
