Neutral Lipid Assay

A neutral lipid assay is a laboratory method for detecting and quantifying nonpolar storage lipids, including triglycerides and cholesteryl esters, in biological or environmental samples. The assay typically separates neutral lipids from more polar cellular components and measures them through dye-based fluorescence, absorbance, or chemical extraction, with signal intensity related to lipid abundance. In environmental research, these measurements help assess energy storage in organisms, evaluate physiological responses to pollutants or changing conditions, and compare lipid accumulation across species, tissues, or habitats. Neutral lipid profiles can therefore provide evidence of organismal health, ecological stress, and alterations in carbon and energy allocation.

A simple protocol to determine the neutral lipid content of algal cells using a Nile Red staining procedure is described. This time-saving technique offers an alternative to traditional gravimetric-based lipid quantification protocols. It has been designed for the specific application of monitoring bioprocess performance.

This study describes an imaging-based micro-neutralization assay to analyze the antigenic relationships between viruses. The protocol employs a flatbed scanner and has four steps, including titration, titration quantitation, neutralization, and neutralization quantitation. The assay works well with current circulating influenza A(H1N1)pdm09, A(H3N2), and B viruses.

In this video, we demonstrate in vitro staining of neutral lipids in hepatocytes to detect hepatic steatosis or excessive fat accumulation in liver. Excessive fat is accumulated in the cells in form of lipid droplets. Oil Red O is a lipid soluble dye that binds to intracellular neutral lipids in these lipid droplets. When observed under a light microscope, these intracellular lipid droplets appear intensely red in color.
