Aphid Culturing

Aphid culturing is the controlled maintenance and reproduction of aphid colonies under laboratory conditions, providing consistent biological material for genetic research. Researchers regulate host plants, temperature, humidity, and photoperiod while isolating or tracking individual lineages; many aphids reproduce parthenogenetically, allowing clonal populations to be maintained and compared. Cultured aphids support studies of heredity, phenotypic variation, host adaptation, insect-plant interactions, and symbiosis. Standardized colonies also improve the reproducibility of experiments by reducing environmental variation and enabling researchers to examine how genetic differences influence development, reproduction, and responses to changing conditions.

Aphid microinjection of virus, the procedure we will show you today, is a technique allowing researchers to inject virus directly into the hemocoel of the aphid, bypassing the gut, one of the 2 major barriers for virus transmission in a circulative manner. The same technique is also used to inject dsRNA for RNAi.
