Aav Production

AAV production is the laboratory process of generating and purifying adeno-associated virus (AAV) vectors, which can deliver genetic material to cells for research and therapeutic development. It typically involves introducing a vector genome together with rep and cap genes and helper functions into producer cells, enabling viral capsid assembly, genome packaging, and release; downstream clarification and purification enrich the desired vector. In immunology and infection research, AAV production supports controlled delivery of genes, antigens, or immune-modulating sequences to study host responses and pathogen biology, while scalable, consistent manufacturing improves reproducibility and advances preclinical evaluation of gene-based interventions.

We demonstrate the basic technique to molecularly engineer and evolve synthetic Adeno-associated viral (AAV) gene therapy vectors via DNA family shuffling. Moreover, we provide general guidelines and representative examples for selection and analysis of individual chimeric capsids with enhanced properties on target cells in culture or in mice.

In this protocol, AAV2 vector is produced by co-culturing Spodoptera frugiperda (Sf9) insect cells with baculovirus (BV)-AAV2-green fluorescent protein (GFP) or therapeutic gene and BV-AAV2-rep-cap infected Sf9 cells in suspension culture. AAV particles are released from the cells using detergent, clarified, purified by affinity column chromatography, and concentrated by tangential flow filtration.

In this video, we demonstrate an in vitro colorimetric assay to detect the presence of neutralizing antibodies against alkaline phosphatase-expressing recombinant adeno-associated virus serotype 6 (rAAV6) vectors in sheep serum samples. The assay utilizes the reaction between the AAV encoding a human placental alkaline phosphatase gene and its substrate to generate an insoluble purple product.
