Antibody-mediated Phagocytosis

Antibody-mediated phagocytosis is an immune process in which antibodies tag microbes or abnormal cells for engulfment and destruction by phagocytes, linking adaptive and innate immunity. Antibodies bind antigens on a target’s surface, while their exposed Fc regions engage Fc receptors on macrophages, neutrophils, and other phagocytic cells; this receptor signaling promotes target attachment, internalization into a phagosome, and degradation after fusion with lysosomes. In immunology and infection, the process helps clear extracellular bacteria, fungi, and infected cells, while also shaping inflammation and antigen presentation. Its principles support vaccine evaluation, studies of host-pathogen interactions, and antibody-based therapies that recruit immune cells against tumors or infectious agents.

This video demonstrates an assay to measure the antibody-dependent cellular phagocytosis (ADCP) by breast milk phagocytes. Fluorescently labeled microspheres conjugated to an antigen-antibody complex, are combined with a cell suspension isolated from breast milk to induce antibody-dependent cellular phagocytosis by the phagocytic leukocytes. Upon labeling the cells with antibodies against common leukocyte antigen (CD45), flow cytometry is used to measure the fluorescence intensity of...

Source: Nagre, N., et al. Alveolar Macrophage Phagocytosis and Bacteria Clearance in Mice. J. Vis. Exp. (2019).This video demonstrates the assessment of complement receptor-mediated phagocytosis using sheep red blood cells opsonized with IgM and complement, followed by their uptake by mouse macrophages. The extent of phagocytosis is quantified by measuring a blue chromogenic product formed through hemoglobin-catalyzed oxidation of diaminofluorene.
