Apc Fluorescence

APC fluorescence refers to the red-to-far-red light emitted by allophycocyanin, a phycobiliprotein fluorophore widely used to label antibodies and other probes. When excited by appropriate red light, APC absorbs photon energy and releases it at a longer wavelength, allowing instruments to detect tagged molecules with limited interference from many shorter-wavelength fluorophores. In immunology and infection research, APC-conjugated antibodies support flow cytometric identification and measurement of immune-cell populations, surface markers, cytokine responses, and pathogen-specific signals. Its bright emission and compatibility with multicolor panels make APC useful for characterizing cellular phenotypes and immune responses, although spectral spillover may require compensation.

Quantification of Cytokine-Induced Cell Death in Human Colonic Organoids Using Live Fluorescence Microscopy

2024

This protocol describes a simple and cost-effective method to investigate and quantify cell death in human colonic organoids in response to cytotoxic perturbagens such as cytokines. The approach employs a fluorescent cell death dye (SYTOX Green Nucleic Acid Stain), live fluorescence microscopy, and open-source image analysis software to quantify single-organoid responses to cytotoxic stimuli.

Introduction to Fluorescence Microscopy

2023

Fluorescence microscopy is a very powerful analytical tool that combines the magnifying properties of light microscopy with visualization of fluorescence. Fluorescence is a phenomenon that involves absorbance and emission of a small range of light wavelengths by a fluorescent molecule known as a fluorophore. Fluorescence microscopy is accomplished in conjunction with the basic light microscope by the addition of a powerful light source, specialized filters, and a means of fluorescently...
