Cell Density Separation

Cell density separation is a laboratory technique that isolates cells according to differences in buoyant density, producing enriched populations for analysis and experimentation. In a density-gradient medium, centrifugation generates layers with different densities; cells migrate until they reach the region where the medium matches their buoyant density, allowing fractions to be collected separately. In immunology and infection research, this approach can enrich peripheral blood mononuclear cells, distinguish leukocyte populations, or help separate host cells from microbial or cellular debris before flow cytometry, culture, or molecular assays. Cleaner, more defined samples improve measurements of immune responses, pathogen interactions, and disease-associated cellular changes.

We demonstrate the use of discontinuous density gradients to separate bacterial populations based on capsule production. This method is used to compare capsule amount between cultures, isolate mutants with a specific capsule phenotype, or to identify capsule regulators. Described here is the optimization and running of the assay.

Soil density fractionation separates soil organic matter into distinct pools with differing stabilization mechanisms, chemistries, and turnover times. Sodium polytungstate solutions with specific densities allow the separation of free particulate organic matter and mineral-associated organic matter, resulting in organic matter fractions suitable for describing the soil response to management and climate change.
