Concanavalin A Assay

The Concanavalin A assay is an immunological method that measures lymphocyte activation, most commonly by assessing T-cell proliferation after exposure to the plant lectin Concanavalin A (ConA). ConA binds mannose- and glucose-containing glycans on cell-surface glycoproteins, cross-linking receptors and triggering signaling pathways that promote cytokine production and cell division. Researchers quantify responses using DNA-synthesis, metabolic, or cell-count readouts, often comparing stimulated and unstimulated cultures. In immunology and infection research, the assay helps evaluate cellular immune function, host responses to pathogens, immunosuppression, and the effects of drugs or microbial products on T-cell activity.

Upon the addition of bile salts in the culture of a pathogenic enteric bacteria, biofilm production is detected using a fluorescent lectin that binds to the polysaccharides in the extracellular polymeric substance (EPS) matrix of the biofilm.

A Rabbit Model of Aqueous-Deficient Dry Eye Disease Induced by Concanavalin A Injection into the Lacrimal Glands: Application to Drug Efficacy Studies

This article describes the development of a method to induce acute or chronic dry eye disease in rabbits by injecting concanavalin A to all portions of the orbital lacrimal gland system. This method, superior to those already reported, generates a reproducible, stable model of dry eye suitable for the study of pharmacological agents.
