Immature Dcs

Immature dendritic cells (DCs) are sentinel antigen-presenting cells that continuously sample tissues for signs of infection or damage before initiating adaptive immune responses. They efficiently capture pathogens and soluble antigens through phagocytosis, macropinocytosis, and receptor-mediated endocytosis, while expressing relatively low levels of major histocompatibility complex class II and costimulatory molecules. Recognition of pathogen-associated or damage-associated signals triggers DC maturation, antigen processing, increased surface presentation, and migration to draining lymph nodes. There, mature DCs activate naïve T cells and help shape immune responses, making immature DCs essential for pathogen surveillance, immune tolerance, vaccine design, and studies of immunopathology.

Generation of Immature, Mature, and Tolerogenic Dendritic Cells from Monocytes

2025

This video demonstrates the generation of immature, mature, and tolerogenic dendritic cells, or DCs, from human monocytes. The incubation of monocytes with growth factors and cytokines leads to the production of immature DCs. In response to immune regulatory molecules, immature DCs differentiate into tolerogenic and mature DCs.

Generation of Immature, Mature and Tolerogenic Dendritic Cells with Differing Metabolic Phenotypes

Immature dendritic cells can be selectively differentiated into tolerogenic or mature dendritic cells to regulate the balance between immunity and tolerance. This work presents a means to generate from immature monocyte derived dendritic cells (moDCs), in vitro tolerogenic and mature moDCs that differ in metabolic phenotypes.

Rearing Ixodes scapularis, the Black-legged Tick: Feeding Immature Stages on Mice

The production of healthy laboratory-reared ticks is essential to studies on tick biology, and tick-pathogen interactions. Here we demonstrate a simple protocol for immature tick feeding that is cost-effective and less stressful to mice.

siRNA-Based Inhibition of Autophagy in Herpes Simplex Virus-Infected Immature Dendritic Cells

2025

This video demonstrates a small interfering RNA (siRNA)-based technique to interfere with autophagic flux. A siRNA targeting FIP200 mRNA is introduced into monocyte-derived immature dendritic cells (iDCs) using electroporation, causing the knockdown of FIP200 expression and leading to the inhibition of nuclear lamin disruption via autophagy. Upon adding herpes simplex virus (HSV), the nuclear egress of viral progeny is inhibited due to the absence of lamin disruption via autophagic flux.
