Lithium Acetate Transformation

Lithium acetate transformation is a laboratory method for introducing exogenous DNA into yeast cells, enabling researchers to modify their genetic material and study gene function. In this process, lithium acetate helps increase cell-wall and membrane permeability, while polyethylene glycol promotes contact between DNA and the cell surface; a heat-shock step can further support DNA uptake, followed by selection of successfully transformed cells. The method provides a practical route for generating engineered yeast strains used to express proteins, investigate cellular responses, and develop experimental systems relevant to immunology and infection research. Its simplicity and adaptability make it valuable for routine molecular genetics.

2023

S. cerevisiae are unicellular eukaryotes that are a commonly-used model organism in biological research. In the course of their work, yeast researchers rely upon the fundamental technique of transformation (the uptake of foreign DNA by the cell) to control gene expression, induce genetic deletions, express recombinant proteins, and label subcellular structures. This video provides an overview of how and why yeast transformation is carried out in the lab. The important features of yeast...

A method for the determination of acetate kinase activity is described. This assay utilizes a direct reaction for determining enzyme activity and kinetics of acetate kinase in the acetate-forming direction with different phosphoryl acceptors. Furthermore, this method can be utilized for assaying other acetyl phosphate or acetyl-CoA utilizing enzymes.

Solid-state Graft Copolymer Electrolytes for Lithium Battery Applications

2013

Lithium ion batteries employ flammable and volatile organic electrolytes that are suitable for ambient temperature applications. A safer alternative to organic electrolytes are solid polymer batteries. Solid polymer batteries operate safely at high temperatures (>120 °C), thus making them applicable to high temperature applications such as deep oil drilling and hybrid electric vehicles. This paper will discuss (a) the polymer synthesis, (b) the polymer conduction mechanism, and (c) provide...
