Luciferase-expressing Tumor Cells

Luciferase-expressing tumor cells are cancer cells genetically modified to produce luciferase, an enzyme that enables noninvasive measurement of tumor growth and distribution. After administration of a compatible substrate, luciferase catalyzes a light-producing reaction, and sensitive imaging systems detect the emitted bioluminescence as a signal proportional to viable labeled-cell numbers under defined conditions. In immunology and infection research, these cells support longitudinal studies of tumor progression, immune-cell activity, treatment response, and pathogen-associated effects in living models. By tracking the same tumor over time, researchers can reduce sampling requirements and relate changes in bioluminescent signal to disease burden and therapeutic outcomes.

Circadian clocks function within individual cells, i.e., they are cell-autonomous. Here, we describe methods for generating cell-autonomous clock models using non-invasive, luciferase-based real-time bioluminescence technology. Reporter cells provide tractable, functional model systems for studying circadian biology.

A method for reliably grafting luciferase-tagged human malignant peripheral nerve sheath tumor cells into the sciatic nerve of immunodeficient mice is described. The use of bioluminescence imaging to demonstrate proper establishment of tumor grafts and criteria for random segregation of animals into study groups are also discussed.
