Mouse Bone Marrow Isolation

Mouse bone marrow isolation is a laboratory technique for recovering hematopoietic and immune cells from the marrow cavities of mouse bones, providing a primary source for studying blood and immune development. The procedure typically involves aseptic removal of long bones, flushing the marrow with a buffered medium, dispersing cell clumps, and filtering or centrifuging the suspension to obtain viable cells. Isolated marrow can support flow cytometry, cell culture, differentiation assays, transplantation studies, and infection models. In immunology and infection research, this preparation enables analysis of progenitor populations, myeloid and lymphoid responses, and how pathogens or inflammatory signals alter hematopoiesis.

We describe a protocol for isolation and purification of neutrophils from mouse bone marrow by density gradient centrifugation and for neutrophil labeling using CellTracker dyes. This represents a simple, fast, reproducible and economical method for obtaining large numbers of neutrophils for downstream functional studies or adoptive transfer and tracking experiments.
