Engulfment Analysis

Engulfment analysis is a method for measuring how efficiently immune cells recognize, bind, and internalize particles, microbes, or target cells, providing a quantitative view of phagocytic activity. During engulfment, receptors on phagocytes trigger actin cytoskeletal remodeling that extends the cell membrane around the target and closes it within a phagosome, while experimental readouts distinguish internalized material from surface-bound material. In immunology and infection research, this analysis helps assess macrophage and neutrophil function, compare pathogen uptake, evaluate host defense mechanisms, and determine how treatments or microbial factors alter cellular clearance.

Experimental Analysis of Apoptotic Thymocyte Engulfment by Macrophages

Here, we present a protocol to prepare apoptotic thymocytes and peritoneal macrophages and analyze the efficiency of efferocytosis and the specific inhibitor-mediated blocking of apoptotic thymocytes engulfment. This protocol has a broad application in cell-mediated clearance of other particles including artificial beads and bacteria.

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Results demonstrate that high content analysis represents an exciting novel technology for neurotoxicity assessment.
