Plasmodesmata Localization

Plasmodesmata localization is the identification and spatial mapping of plasmodesmata, membrane-lined channels that connect neighboring plant cells and coordinate intercellular communication. These junctions contain a plasma-membrane-lined pore and an endoplasmic reticulum-derived desmotubule, while callose deposition can narrow the channel and regulate molecular movement. Fluorescent labeling of plasmodesmal proteins and microscopy can reveal their distribution, abundance, and changes during infection. In plant immunity research, localization studies help determine how pathogens exploit or alter cell-to-cell connectivity, how defense signals spread through tissues, and how changes in plasmodesmal permeability influence infection outcomes.

Plant intercellular connections, the plasmodesmata (Pd), play central roles in plant physiology and plant-virus interactions. Critical to Pd transport are sorting signals that direct proteins to Pd. However, our knowledge about these sequences is still in its infancy. We describe a strategy to identify Pd localization signals in Pd-targeted proteins.

Nano-fEM: Protein Localization Using Photo-activated Localization Microscopy and Electron Microscopy

We describe a method to localize fluorescently tagged proteins in electron micrographs. Fluorescence is first localized using photo-activated localization microscopy on ultrathin sections. These images are then aligned to electron micrographs of the same section.
