Blood Plasma Isolation

Blood plasma isolation is the laboratory process of separating the liquid component of blood from cells, producing a sample that contains water, proteins, electrolytes, hormones, and other circulating molecules for medical analysis and treatment. Typically, blood is collected into a tube containing an anticoagulant and centrifuged, causing denser red and white blood cells and platelets to sediment while plasma remains in the upper layer; careful pipetting then transfers the plasma without disturbing the cell fraction. In medicine, isolated plasma supports biochemical testing, biomarker measurement, coagulation studies, drug monitoring, and preparation of plasma-derived products, making consistent handling essential for reliable results.

Composition of Blood Plasma

2025

Blood plasma is a fluid that contains approximately 92% water and 8% solutes. The solutes include various types of proteins, which constitute about 7% of the total solutes in the plasma. The high-molecular-weight proteins—albumins, globulins, and fibrinogen—are essential to plasma function. Albumins, making up about 60% of the plasma proteins, maintain the osmotic balance within blood vessels by preventing excessive water leakage. Additionally, albumins serve as carrier proteins, binding to...

Extracellular vesicles present in blood have been suggested as novel biomarkers for various diseases. Here, we present a protocol for the isolation of large plasma membrane-derived microvesicles from peripheral blood samples and their subsequent analysis by conventional flow cytometry and Western Blotting.
